Although there is no obvious evidence of image duplication in the revised Fig. S8 posted on the authors' website (https://liulab.seas.harvard.edu/sites/projects.iq.harvard.edu/files/jialiulab/files/supplementary_information.pdf), further concerns have emerged when comparing the published Fig. S8 with the corrected Fig. S8, as detailed below:
It is now evident that the original miniSOG+/monomer-/irradiation- image has been repurposed as the miniSOG+/monomer-/irradiation+ sample (highlighted in red boxes). Additionally, the original miniSOG+/PDAB+/irradiation+ image now serves as the miniSOG-/PANI+/irradiation+ sample (indicated by magenta boxes), while the original miniSOG-/PANI+/irradiation+ image has been reassigned as the miniSOG+/PDAB+/irradiation+ image (denoted by green boxes).
This pattern suggests that the authors may have had several negative control images, which they seemingly assigned to these negative control groups in an arbitrary manner.

Regarding Figure 5, the authors provide the data for time series measurements in single neurons. However, it appears there are some inconsistencies in the number of neurons between each time point. It is possible some of these datapoints are completely obscured by others within each plot, or have been omitted entirely. May the authors provide the raw data for this figure and clarify if specific timepoints were omitted?
In panel 5E, the “miniSOG- PANI+” condition contain 5 datapoints at the “Before” condition and 4 datapoints at the 6 min condition. However, there appears a neuron denoted by black at the 7 min timepoint that appears once (behind the red datapoint), and a neuron denoted by yellow only in the beginning and final timepoints.
In panel 5G, the miniSOG+ PDAB+ condition have 4 datapoints in the “before” condition and 5 datapoints in the subsequent timepoints. Additionally, if individual colors denote individual neurons, there is the purple datapoint at 6 min. which takes the place of the neuron denoted by green.
In panel 5H, the miniSOG- PDAB+ condition contain 5 datapoints at the “Before” and 5 min. conditions but 6 datapoints at following timepoints. Additionally, the miniSOG+ PDAB+ condition have 6 datapoints at the “Before” and “7 min” conditions, but 4 datapoints at the 5 min. and 5 datapoints at the 6 min. conditions.
In panel 5I, the miniSOG+ PANI+ condition appears to be missing the “black” datapoint at the 6 min. timepoint.
Could the authors provide the raw data to clarify if: (1) the missing datapoints are perfectly obscured by other datapoints in the graphs, or if (2) data from specific neurons were omitted and/or mixed and matched. If data was omitted or mixed and matched in the time series, it is then reasonable to explain as to why this was done.
